Spent way too long looking in the wrong places for this.
After updating my Roku Ultra to 14.1.4, my Harmony remote started behaving very badly with the Roku. My experience was extremely delayed keypresses, others have reported a complete failure to function. Solution: 14.1.4 introduced a new setting, under Advanced System Settings / Control with Mobile Apps. The default setting is "Limited", which really... well, limits control with mobile apps. Change it to "Permissive" and everything works again just like before. Hopefully this will save someone else the trouble of diving into the twin hellscapes of Logitech and Roku support. EDIT: It appears the problem may be model-specific. I have another, more recent, Ultra, and it didn't even have a "Limited" option. It defaulted to "Default" (which wasn't an option on my older Ultra), which seemed to be fine. I changed it to "Permissive" too out of an abundance of caution for future updates.
