That isn't to say that I don't like Holy Grail. It is a great film, with some great, eminently quotable, dialogue - "I fart in your general direction. Your mother was a hamster and your father smelt of elderberries." Oh, and who can forget The Holy Hand-grenade of the Antioch...
Could not have said it any better myself.

Even though I know that Holy Grail will trump in this poll, Life Of Brian is by far the best in my opinion. It's far less reliant on the usage of "silliness" for laughter. And, what "silliness" IS used often has some cerebral context to back it up... aside from the alien intermission bit. I still scratch my head everytime I see that part.

I sometimes wonder if they threw that whole alien sequence as a piss-take of the 'god was an alien' theory. Or, a bit of a dig at the 'I was abducted by aliens' brigade. Of course, it could just simply be for a bit of fun...

My gut first wanted to go with Grail for the win. I really enjoyed Grail (and Life for that matter), but as I read the thoughts here, and think about which is actually the better movie, I have to go with Brian as well; although it has fewer uncontrollable laughing-out-loud moments, I think it's just a smarter, better product overall.

+1 The Holy Grail has some really funny bits (i.e. weighing a witch) but IMO it's the least mature of the three. It's probably the most accessible, so it's the crowd pleaser.

Talking

I'm a huge Monty Python fan. However, some of their skits are not that funny to me. I'm talking about Flying Circus. Some skits are so funny I am on the floor with hilarity! For me this attitude serves me well when I watch one of the troupes featured films. I find it interesting that the troupe all went to college in England and were required to take about 21 hours of philosophy. A large portion of their skits are based upon these studies. I majored in philosophy so I find it really funny!

Oh yea. Meaning of Life for me.

I much prefer http://www.youtube.com/watch?v=WlBiLNN1NhQ to every Holy Grail tune. It's a far more dominant "catchy" tune to anything else Python has done musically.
