Few moments in film history have been as satisfying as that one.

Also, the ending for Saw.
Cap lists the hammer in the comics. I was annoyed when he didn’t in Age of Ultron. I was seriously bummed when the hammer was destroyed in Ragnarock. I then got excited when Thor collects the hammer from the past. For me at that point, Cap HAD to pick up the hammer.

An amazing reveal in Saw but that scene was spoiled for me but I’m glad as I would never have seen the movie without being spoiled.

Everyone seems to have various criteria for what the OP is asking, but I’ll play anyway.

Rapturous crowd reactions all 3x I saw it on opening weekend.

I remember going to see the Simpsons and as homer blacks out and the screen blacks out the theater has a black out. It really confused everyone when the screen did not come back on again.

Can’t believe no one has mentioned the original chest bursted in Alien yet! That was quite a wallop on its original release.

Or the cross scene in The Exorcist.

The car hit a bump scene in Pulp Fiction. The whole theatre was laughing hysterically.

The reaction to Andrew Garfield was wild and completely unexpected, but Maguire's reveal was biggest reaction I have ever experienced in a cinema. This was the first screening on opening day, and they went absolutely ape-shit!

Very few films rate a response at my local seaside independent 3-screener, but this one blew the roof off! Cheers, applause and even some foot-stomping, all of which took about a minute to die down.

It'll remain a very pleasant memory.

I can’t pick just one, but off the top of my head, these moments had strong audience reactions:

Yoda unveiling his lightsaber in Attack of the Clones (cheering/clapping)

The death reveal in Bridge to Terabithia (I’ve never heard an audience collectively gasp before)

The trailer attack in The Hills Have Eyes (2006) (stunned silence)

The silence in the water during Titanic (sniffling to outright sobbing)

Pennywise grabbing Beverly in IT Ch. 1 (several people literally leapt out of their seats)

Hero battlefield reveals in Endgame (eruption of cheers/clapping)
