Bela Lugosi turned down Frankenstein.

He was a huge star right after starring in Dracula, and Universal wanted him to play the monster -- but he turned them down because of the heavy make-up and lack of dialogue. The role went to the then unknown Boris Karloff who became a sensation and quickly eclipsed Lugosi as Hollywood's biggest horror star. He went on to major success as Bela drifted into lesser film roles and faded away

Many film historians consider that the biggest mistake of Lugosi's career.

Overall it does not change anything. These role being turn down or having to drop because of conflict in schedule and what not have no meaning to these actors, they have only meaning to the media, who love to bring this back up every once an a while.

Would LOTR been better with Connery as Gandalf? Would we even care about Indy today if Tom Selleck had been Indy in Raiders? Would The Matrix had been as popular with Will Smith? All of these movies might have been nothing more then obscure reference now if the actors who did them had not been in them. It truly mean's nothing at all who refuse or could not do a role. Seriously anyone think Al Pacino had been such a great choice for Han Solo? Beside every actor of a certain age made audition for the role back in 1975. It just fun to talk about it but that's about it.

No, Selleck did not turn down Indiana Jones. As mentioned earlier, he was the first choice for the role, but he had just committed to do Magnum PI and the producers would not release him from his contract.
