Yes, and the only way to really know where a sub gives the flattest response is to measure the room as you experiment. Anything else, especially relying on playing music, is just guessing. Room peaks and nulls occur at specific frequencies, but music plays only a few different bass frequencies that depends on the musical key of the piece. You could spend an hour playing many types of music, and hope that your ears are accurate enough to know what sounds right. Versus measuring that takes only ten seconds per measurement.
