Noise reducing when used during film xfers for the most part will reduce the amount of visible grain in the picture. Especially for 16mm neg when they didn't intend to shoot it like that...alot of people miss the course on how to properly light a scene since it can be fixed in the post production process most of the time.

It's a subjective call, depending on what you like. If you want the most faithful experience to the film, leave any forms of noise reduction off. If you don't mind sacrificing a little detail for a more sanitized look, turn it on. The level will depend on the film. Some films are grainier than others. Though DNR was really intended more for compression related noise in poor encodings than natural film grain. Not all DNR algorithms work very well either. Some hardly do anything at all. And some will soften the picture more than others. DNR can also add artifacts and video anomolies all its own.

Personally, I never use such features and don't recommend them. They were more worthwhile with DVD, but rarely does a BD come out with consistent enough problems to justify noise reduction. But if you do, the decision whether to activate DNR and to what extreme it's applied needs to be made on a title by title basis.
